چکیده

A disposable sensor has been developed for the measurement of copper(II) concentration in aqueous solution based on a change in the fluorescence of porphyrazine 2,7,12,17-tetra-tert-butyl-5,10,15,20-tetraaza-21H,23H-porphine (TP). The sensor was constructed by spin-coating a polyester support with a PVC solution containing TP, a plasticizer, the chelating agent Zincon and the ion-pairing benzetonium chloride. The measurement principle is based on the radiationless resonance energy transfer (RET) from TP immobilized in membrane, and acting as fluorescence donor, to Zincon acting as an acceptor induced by copper(II). The absorption spectrum of the Zincon–Cu(II) complex presents adequate overlapping with the emission spectrum of TP, producing a useful analytical signal by the RET process. The disposable sensor responds to copper(II) irreversibly over a dynamic range from 0.039 to 14 mol L−1 (2.5–890 g L−1) with a sensor-tos T s ©

Ratiometric fluorescent sensor proteins with subnanomolar affinity for Zn(II) based on copper chaperone domains.

The ability to image the concentration of transition metals in living cells in real time is important for further understanding of transition metal homeostasis and its involvement in diseases. The goal of this study was to develop a genetically encoded FRET-based sensor for copper(I) based on the copper-induced dimerization of two copper binding domains involved in human copper homeostasis, Ato...

Nucleic Acid Sandwich Hybridization Assay with Quantum Dot-Induced Fluorescence Resonance Energy Transfer for Pathogen Detection

This paper reports a nucleic acid sandwich hybridization assay with a quantum dot (QD)-induced fluorescence resonance energy transfer (FRET) reporter system. Two label-free hemagglutinin H5 sequences (60-mer DNA and 630-nt cDNA fragment) of avian influenza viruses were used as the targets in this work. Two oligonucleotides (16 mers and 18 mers) that specifically recognize two separate but neigh...

Robust, specific ratiometric biosensing using a copper-free clicked quantum dot-DNA aptamer sensor

(2013) Robust, specific ratiometric biosensing using a copper-free clicked quantum dot-DNA aptamer sensor. 5 We report herein the successful preparation of a compact, functional CdSe/ZnS core/shell quantum dot (QD)-DNA conjugate via the highly efficient copper-free " click chemistry " (CFCC) between a dihydro-lipoic acid-polyethylene glycol-azide (DHLA-PEG-N 3) capped QD and a cyclooctyne modif...

A single-bead telomere sensor based on fluorescence resonance energy transfer.

We present a 200 nm in-diameter single-bead sensor for the detection of single, unlabeled DNA molecules in solution using fluorescence resonance energy transfer technology. DNA-bound Alexa 488 and Crimson 625 loaded on commercial beads served as the donor and acceptor, respectively. Binding of the target DNA to the single bead sensor induces G-quadruplex stretching, resulting in a decrease in f...

His-tags as Zn(II) binding motifs in a protein-based fluorescent sensor.

Fluorescent indicators that allow real-time imaging of Zn(II) in living cells are invaluable tools for understanding Zn(II) homeostasis. Genetically encoded sensors based on fluorescence resonance energy transfer between fluorescent protein domains have important advantages over synthetic probes. We discovered that hexahistidine tags have a strong tendency to dimerize upon binding of Zn(II) in ...
